Easiest way to get EVERY character:

Beat the Subspace Emissary. You'll have a little bit of fun while you're doing
it, too.

Once you've beaten it, you'll have unlocked most characters. There's a few
remaining ones, however. You can also get these in Subspace Emissary as well,
but they're only available after you've beaten the game.

Toon Link - In "The Forest", enter the first door hovering in the air.
Easy as cake.
Wolf - In "The Ruins", proceed as normal until you're riding an
elevator downward and then avoiding spikes on the walls. Instead of taking the
first stop, continue to go all the way to the bottom of this shaft and go in
the door there.
Jigglypuff - I found this door the hardest to reach. It's in "The
Swamp", and it's pretty much right after the first barrel launcher. It's
difficult to get into, just avoid the barrels and keep in mind you can swim.

If you don't want to do SSE, you're out of luck as far as I'm concerned, but
here's some alternate ways to earn SOME characters (there's others but I don't
know them):

Captain Falcon - Clear Classic mode on Normal difficulty in 12 minutes.
Falco - Clear 100-Man Brawl
Ganondorf - Clear Classic mode on Hard difficulty as Link or Zelda; or
battle 200 times

You can also unlock EVERY character by playing enough brawls. We're talking
in the thousands, though, to get every character that way.

Here's how to unlock stages (that don't automatically come with unlocked
characters):

75 m - Use DK 20 times in brawls
Big Blue (Melee) - Use Captain Falcon 10 times in brawls
Green Greens (Melee) - Use Kirby 20 times in brawls
Hanenbow - Clear Event #28
Jungle Japes (Melee) - Play Melee stages 10 times
Luigi's Mansion - Use Luigi three times in brawls.
Mario Bros - Clear Event #19
Pokemon Stadium (Melee) - Brawl on the Pokemon Stadium 2 stage 10 times
Spear Pillar - Clear Event #25

Here's how to unlock events:

All-Star Mode - Unlock all characters
Boss Battle mode - Clear Adventure and Classic
Co-op events #9-13 - Complete the Subspace stage in Adventure mode
Co-op Events #14-20 - Clear 10 Co-op events and unlock all chars
Co-op Event #21 - Clear Co-op events #1-20
Events #11-20 - Clear 7 events
Events #21-28 - Complete the Subspace stage in Adventure mode
Events #29-40 - Unlock all chars
Event #41 - Clear Events #1-40

You can unlock a level of Target Smash for completing Classic mode on every
difficulty level.

And lastly, Assist Trophies:

Barabara - Unlock 25 songs.
Custom Robo - Play in 100 brawls
Gray Fox - Have Snake join you in the Subspace Emissary
Infantry and Tanks - Play in 300 brawls
Isaac - Play in 200 brawls
Shadow The Hedgehog - Have Sonic join you in the Subspace Emissary
