Can i run the internal updater from operausb 10 to 10.01 or should i update the old manual way by copying the files described here: http://my.opera.com/opera-usb/forums/topic.dml?id=127989 ?

making snapshots into USB versions is a waste of time. When Opera 10.10 *Final* comes out then a USB version of it will follow as well
